I am in a founderless nation. How do I take control and lock my region?

Become the delegate by getting two endorsements, and being the most endorsed.

Get another player's nation that's present there to join the WA and endorse you (although I seem to recall that that Vandoosa-the-player is an active participant in the raiding side of things...), or another player to move their WA nation in and endorse you, so that you become delegate. Then keep on accumulating 'Influence' until you've got enough to remove the current Regional Officers and then to password the region using an "invisible" password (although that costs more Influence than a visible) so that any raider spies present can't see it.
Note, though, that that would leave the region inaccessible to potential recruits.

Looking at the current situation, however, it would probably be much more practical for you to copy all of the region's current details that you want to preserve (original founder's name, History, badges, RMB, whatever...), found a new region with a very similar name and with executive authority for yourself there as Founder, move into that region, and post all of the copied data about the older one in factbooks or dispatches attached to the new one's World Factbook Entry.

One endorsement is enough for this, if nobody else there has any: Two is only the minimum for submitting WA proposals.

Well, first you'll need to find out if there is already currently a WA Delegate. If there is, you'll need to get at least one more Endorsement than they have, and then you'll need to wait until the WA Update to become Delegate.

If there is not already a WA Delegate of the Region, you'll need at least 1 Endorsement to become the WA Delegate of the Region, but you'll need at least 2 to submit Proposals. You can then do as you please once you become WA Delegate (Executive) of your Region.

You may have a problem. Lily seems to have already passworded the region. Unless you know the password, no one else is getting in to endorse you.

Yes, they were the one who passworded it. Didn’t remove the dispatches while they were delegate, but the OP was the one to password the region.
