Purpose of Idea:

It is possible to genetically engineer bacteria capable of generating 1,2,4-butanetriol, a chemical precursor to the explosive BTTN, using common glucose (a major energy source for eukaryotic cells) as a substrate. Nitration, an essential step in converting 1,2,4-butanetriol to BTTN, is known to happen in a biological context in a variety of substrate molecules, including lipids and tyrosine amino acids, and we believe it is feasible to engineer a pathway to nitration of 1,2,4-butanetriol as well.

The biology of braconid and ichneumon wasps and the bracoviruses and polydnaviruses they produce provide a template for the use of genomically encoded viruses to enact trans-species viral infection. Direct modification of the human genome (e.g. through vectored immunoprophylaxis) is a potential strategy in the control of major pandemic diseases including HIV.

We propose to create a DNA-based drug to be administered parenterally (intramuscular route) to human subjects in regions of endemic arthropod-borne diseases. This drug will cause the subjects' cells to create a virus capable of infecting biting arthropods (with special emphasis on mosquitoes but potentially encompassing ticks and fleas, and also botflies because we hate them), but not infecting humans. The infected arthropods will generate substantial quantities of BTTN explosive in their bodies, and will eventually go out with a bang.
