For the sake of progression, I don't think so. Rache senses people through their emotions, I don't see why the emotions wouldn't remain pure in their expression despite what is drawing them out.

There was some controversy over casting Johansen because a few people thought it was whitewashing and that they should have cast an Asian actress. In response to this, a group decided to do a small social experiment where they stood on the streets of Tokyo and asked pedestrians to share their thoughts on a picture of Johansen dressed as the character. The reactions were overwhelmingly positive, many remarking that she looked beautiful and perfect for the role. When asked whether or not they found it offensive that she wasn't Asian, most of them said no and looked really confused at the question. Then one guy outright said: "Well, anime characters don't typically look Asian, do they?"

Whether the character herself is supposed to be Asian is fairly important as well, and "Well, anime characters don't typically look Asian, do they?" belies the fact that while Anime might not have characters appear Asian as a stylistic choice, the characters themselves may be supposed to be Asian.
Judging from the name, at least, and the setting, the Major seems to lean Asian descent.
Whether the film will be good is, of course, totally independent of this, but I can see why some fans would be a little pissed at the casting choice. Personally, I would like to see more Asians break out in Hollywood, but it's not like Scarlet Johansen is a terrible actress for the role.
