Rio - La Paz - Lima.

Technically Panama only splits North America. And you could also build a McMurdo - Amundsen-Scott - Sanae IV, splitting Antarctica. :p

It would be interesting to see if there is a serious expansion and improvement of the canal to allow bigger cargo ships in and out. That would surely change the geopolitics in the Mediterranean.
