Not necessarily. We could get to alpha centauri before the end of the century, it would just have to be a generation ship because of the hundred year roundtrip.

We have no evidence of races 60 million years more advanced than us.
If there is more-advanced life out there, it's either so far away we'll never find it, deliberately avoiding us, or it's been and gone... probably millions of years ago.
And since there's no evidence at all of more-advanced aliens, the most logical assumption is that any life that IS out there (if any), is less advanced than us. (After all, it's statistically VERY unlikely it would be within thousands of years of our level).

I don't see why we'd need a moonbase. We're not running out of materials that urgently, and if we assemble in space we don't need to deal with even the moon's gravity.

Because of the gravitational well of Earth. How are you getting the stuff (and mind you, it takes a FUCKLOT of stuff to build a viable generational ship) into Earth orbit? It's cheaper to go from the Moon to LEO than from Earth to LEO.
